Mazdutide is a once-weekly injectable dual agonist of the GLP-1 and glucagon (GCG) receptors, primarily used for long-term weight management and the treatment of type 2 diabetes. It was developed by Eli Lilly and licensed to Innovent Biologics, gaining formal approval in China.
How It Works
Unlike standard GLP-1 medications, mazdutide (IBI362) mimics oxyntomodulin. By activating both receptors simultaneously, it:
Suppresses appetite and delays gastric emptying (GLP-1 pathway).
Increases energy expenditure and promotes the reduction of visceral fat (Glucagon pathway).
Clinical Efficacy & Weight Loss
Clinical trials in Chinese adults with obesity have shown impressive, clinically meaningful results. For instance, the Phase III GLORY-1 trials and subsequent studies with higher 9-mg dosages demonstrated average placebo-subtracted weight reductions ranging from 13% to over 15% after 32 to 60 weeks of treatment.
Approved Indications
Chronic Weight Management: Approved in China for adults with a Body Mass Index (BMI) of 28 kg/m² or greater, or 24 kg/m² or greater when accompanied by at least one weight-related comorbidity.
Type 2 Diabetes: Approved in China as a glycemic control agent, delivering reductions in hemoglobin A1c (HbA1c) and fasting plasma glucose.
Side Effects
As with other incretin-based therapies, the most common adverse reactions are gastrointestinal in nature. These are generally mild to moderate and include:
